I caught several last night and while reading up on them saw a couple random post that they sometimes can be poly. Is this actually true and if so has anyone had experience with a poly colony of L. americanus?

This topic with many species of Lasius is, surprisingly, still somewhat unexplored.

There's a small amount of evidence that supports that microgynes are more likely to have association with polygyny in many cases in many species. We don't know if this is the case for some species, however.

I've seen a colony of  Lasius americanus with 5-6 microgyne queens.

If attempting to introduce two queens prior to egg laying is it something that usually takes several attempt by putting both into a test tube then separating them off and on again until they tolerate each other?

Or

Should I introduce them to each other in a slightly larger container first then reduce the size of the container down a bit? Allowing them to have some space and slowly restrict the space until they tolerate each other then place both in the same test tube?
