As long as it doesn't leak the size is not really that important. Due to the cotton at the end sufficient humidity is assured either way.
If you want to allow your ants to move out of the test tube without removing all the cotton (and drying out the tube) you can just push a drinking straw through the cotton so the ants have a smaller opening.
