When I extracted the Camponotus colony from the piece of wood there was a large abdomen winged ant.  The wings are now removed.  Should I move her and a few workers to a separate test tube?

I doubt she’s fertilized, unfortunately. Camponotus in Alberta are generally monogynous. This is a behaviour that occasionally happens where a reproductive alate within the colony removes her own wings.
